ARA-290
Price range: $60.00 through $70.00
An 11-amino-acid peptide from the helix B surface of erythropoietin, engineered to engage tissue-protective signalling without erythropoietic activity.
Description
ARA-290 is an 11-amino-acid synthetic peptide with the sequence Glp-Glu-Gln-Leu-Glu-Arg-Ala-Leu-Asn-Ser-Ser and a molecular weight of 1257.31 g/mol. It is also known as cibinetide or pHBSP, and derives from a surface region of erythropoietin.
Structural basis
Erythropoietin is a 165-residue glycoprotein best known for stimulating red blood cell production. ARA-290 reproduces a short stretch from the outward-facing surface of its helix B, a region that is not involved in binding the classical erythropoietin receptor.
That distinction is the whole design rationale. Published work describes erythropoietin as signalling through two different routes: the classical homodimeric receptor, which drives erythropoiesis, and a heteromeric receptor combining the erythropoietin receptor with CD131, sometimes called the innate repair receptor, which is associated with tissue-protective signalling. ARA-290 is engineered to engage the second without the first, which is why it is described as non-erythropoietic.
The N-terminal residue is pyroglutamic acid, written Glp. This is a glutamate whose side chain has cyclised onto its own amino terminus, forming a ring. That is deliberate rather than incidental: the closed ring blocks aminopeptidase attack, a common stability strategy in short peptides.
Research background
Published work has examined receptor interaction in cell systems, with attention to the separation between erythropoietic and tissue-protective signalling. Further work has investigated the peptide in rodent models of neuropathy and inflammation.
No regulatory authority has approved ARA-290 for any use. The material supplied here is for laboratory research only, and the majority of published findings remain preclinical.
Storage and handling
Store the lyophilized powder at 36 to 46 °F (2 to 8 °C), desiccated and protected from light. Allow the vial to reach room temperature before opening.
Use 1257.31 g/mol for molar calculations.
Purity and verification
Each lot is analysed by an independent laboratory, with identity confirmed by mass spectrometry and purity by HPLC. Test scope varies by lot; consult the certificate for the lot you receive.
