DSIP

A nine-amino-acid peptide isolated in the 1970s and named for the sleep context of its discovery. No specific receptor has been definitively characterised.
Description
DSIP is a nine-amino-acid peptide with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu and a molecular weight of 848.81 g/mol. Its name stands for delta sleep-inducing peptide, and it is also known as emideltide.
Origin and a caution about the name
DSIP was isolated in the 1970s from the cerebral venous blood of rabbits during induced sleep, and named for the delta-wave activity observed in that experimental setting.
Researchers should treat the name as a historical label rather than a description of established function. The sleep association comes from the circumstances of its discovery, and subsequent work has not produced a settled account of what the peptide does or which receptor, if any, it acts through. No specific DSIP receptor has been definitively characterised. This is unusual among the peptides in this catalogue, most of which have a defined molecular target, and it should shape how experiments are designed and how results are interpreted.
Structure
The sequence is short and unusually rich in small residues: three glycines and two alanines out of nine positions. It carries no cysteine and forms no disulfide bond, and it is not amidated, terminating in a free glutamate. The N-terminal tryptophan is the only bulky residue and is the peptide’s principal chromophore, which makes it the basis for UV detection during analysis and also the reason light protection matters during storage.
Research background
Published work is principally rodent based and considerably older than for most compounds here, with much of the foundational literature dating from the 1970s and 1980s.
The original area of investigation concerned sleep architecture in animal models, with reported observations on electroencephalographic activity.
A second area concerns stress-related endpoints, where published work has examined the peptide in relation to corticotropin and stress-response measures.
A third area concerns observations on thermoregulation and on nociceptive thresholds in animal models.
The literature is notable for inconsistency between studies, and replication has been a recurring difficulty. Researchers should read the primary sources rather than relying on summaries, and should not assume a consistent effect direction. No regulatory authority has approved DSIP for any use.
Storage and handling
Store the lyophilized powder at 36 to 46 °F (2 to 8 °C), desiccated and protected from light. Allow the vial to reach room temperature before opening.
Use 848.81 g/mol for molar calculations.
Purity and verification
Each lot is analysed by an independent laboratory, with identity confirmed by mass spectrometry and purity by HPLC. Test scope varies by lot; consult the certificate for the lot you receive.
