A 44-amino-acid GHRH analogue with an N-terminal hexenoyl group blocking DPP-4 cleavage, studied in growth hormone secretion research.

SKU: TESAMORELIN-PARENT Category:

Description

Tesamorelin is a 44-amino-acid synthetic peptide with a molecular weight of approximately 5136 g/mol. It is an analogue of human growth hormone-releasing hormone, and is also known by the research code TH9507.

Structure

Tesamorelin reproduces the full 44-residue sequence of human GRF(1-44) with one modification: a hex-3-enoyl group attached to the N-terminal tyrosine.

That single addition is the entire point of the molecule. Native GRH is cleaved rapidly by dipeptidyl peptidase-4 at the N-terminus, giving it a circulating half-life measured in minutes. The hexenoyl group obstructs that cleavage site, substantially extending stability without altering the receptor-binding sequence. It is a minimal intervention compared with the lipidation strategies used in the incretin analogues, and it illustrates a different approach to the same stability problem.

Relationship to the other GH compounds in this catalogue

Tesamorelin is a GHRH analogue, meaning it acts at the growth hormone-releasing hormone receptor on pituitary somatotrophs. This is a different receptor from the one engaged by the growth hormone secretagogues such as ipamorelin and hexarelin, which act at the ghrelin receptor.

The two classes are frequently studied together because they engage separate receptors on the same cell population, which is the standard rationale for the GHRH-plus-secretagogue combinations. CJC-1295 without DAC, also in this catalogue, is a GHRH analogue built from the shorter GRF(1-29) fragment with four amino acid substitutions, representing a different engineering approach to the same target.
